ChemikerInnen finden ungewöhnliche Kombination für die umweltschonende Herstellung von beruhigenden und schmerzstillenden Produkten

Der prachtvolle kalifornische Goldmohn und der lästige Schwarzschimmelpilz können eine ungewöhnliche Symbiose eingehen: Wie die Chemiker Wolfgang Kroutil (Uni Graz), Peter Macheroux (TU Graz) und Nick Turner (Uni Manchester) herausgefunden haben, enthalten beide Organismen Biokatalysatoren, die für die Produktion pharmakologisch aktiver Naturstoffe, so genannter Alkaloide, von Bedeutung sind. Die Wissenschafter erprobten in Experimenten, wie die beiden Biokatalysatoren kombiniert werden können - Arbeitsteilung auf molekularer Ebene. Hergestellt wurden dabei Naturprodukte mit beruhigender oder schmerzstillender Wirkung.

Das Enzym aus dem Schimmelpilz kann zwei Formen von Alkaloiden, die sich wie Bild zu Spiegelbild verhalten und unterschiedliche Eigenschaften haben, in nur eine der beiden Formen umwandeln. Der Biokatalysator aus dem Goldmohn kann dieses Produkt in strukturell verwandete chemische Verbindungen umwandeln, die ein erweitertes Wirkspektrum haben.

Den WissenschafterInnen gelang damit die Erzeugung bis dahin unbekannter Alkaloide. Gleichzeitig konnten sie durch die Kombination beider Biokatalysatoren den pharmakologisch wichtigen Naturstoff Scoulerin erstmals im Labor herstellen, und zwar auf höchst effiziente Weise.

„Die Kombination mehrerer Biokatalysatoren in einer einzigen Reaktion hat großes Potenzial und gewinnt immer mehr an Bedeutung. In diesem speziellen Fall konnten wir nicht nur ein völlig neuartiges Reaktionssystem etablieren, sondern wir haben auch zwei Reaktionen kombiniert, die sich mit rein chemischen Mitteln bisher nicht durchführen lassen", betont Wolfgang Kroutil.

Die herausragenden Forschungsergebnisse wurden in der Fachzeitschrift "Angewandte Chemie" publiziert.
