Chemiker lösen Rätsel um Anordnung von Molekül-Kristallen

Molekül-Kristalle bilden sich beispielsweise in Wasser-Eis, kommen in Zucker und Schokolade oder in Medikamenten vor. Deswegen sind ihr Aufbau und ihre Anordnung auch für die Forschung wichtig. Bei einem internationalen Wettbewerb konnten Daniel Boese, Johannes Hoja und ihr Team die Strukturen besonders gut vorhersagen.

Aus welchen Atomen ein Zuckermolekül oder Aspirin besteht, ist hinlänglich bekannt. Wie genau sich die Moleküle dann aber untereinander zu einem Kristall verbinden, kann ganz unterschiedlich sein. „Sie können sich verknäueln, flach übereinanderliegen, in T-Form aneinandergereiht sein – es gibt viele Varianten“, erklärt Daniel Boese. Welche Struktur die stabilste und damit am wahrscheinlichsten zu finden ist, stellt die Wissenschaftler:innen oftmals vor ein Rätsel. Insgesamt gibt es Millionen von verschiedenen Möglichkeiten, die Moleküle anzuordnen. An der Universität Graz wurden Methoden entwickelt, um die Anordnung von Molekülen im Kristall vorhersagen zu können. Das Thema ist unter anderem für die Pharma-Industrie relevant. „Unterschiedliche Strukturen haben auch unterschiedliche Löslichkeiten, zerfallen also beispielsweise schon im Mund oder erst im Magen“, führt Boese aus.

Beste Prognosen

In einem internationalen Wettbewerb des Cambridge Crystallographic Data Centre, der bereits das siebte Mal stattfand, konnten das Grazer Team seine entwickelten Methoden höchst erfolgreich testen. Die 25 teilnehmenden Gruppen aus 20 Ländern mussten aus 500 vorgegebenen Strukturen die jeweils stabilsten herausfinden. „Wir waren die einzigen, denen das in vier von fünf Fällen gelang“, freut sich Boese. Die Ergebnisse des Wettbewerbs wurden nun auch publiziert. Das Interesse an dem wissenschaftlichen Artikel ist groß: So wurde der sechste Wettbewerb bereits 650-mal zitiert – nicht nur seitens der Forschung, auch seitens der Wirtschaft. „Unsere erfolgreichen Vorhersagen am Computer ersparen aufwändige und teure Experimente“, erklärt der Chemiker.
